The soundtrack to the killer Orson Welles-flick by the same name has always been dope as fuck to me-sleazy "detective-flick" fake-jazz/exotica; mancini was a genius IMO.
Herbie Hancock's soundtrack to the Blow-up is a killer too...
Needless to say, I dig most of the classic Blaxploitation soundtracks as well (who doesn't?) with "Superfly" being my favorite and I also really like "Let's Do it again" (yes, I know that movie was more of a comedy) as well as "Trouble man" and the James Brown-ones. "Shaft"-while classic and gamechanging-never struck me as *that* good even if it has some amazing songs; honestly, I like the "three tough guys"-soundtrack more from Isaac and I'm not sure "Truck turner" is worse either...
I'm also a fan of Morricone's spaghetti-western soundtracks (duh!-who isn't?) but I'm having a hard time picking a best one and I don't have any of these in "album"-form, only on Morricone-comps.
I'm also a big fan of italian horror movie-soundtracks but it's the same thing there; I don't really have any albums with the direct soundtrack-just comps; let's just say that Goblin's music to the Dario Argento-flicks is killer. And Fabio Frizzi's soundtracks to Lucio Fulci's City of the living dead and-ESPECIALLY-the Beyond ( http://www.youtube.com/watch?v=acc5PsmHcRg Did the RZA ever sample this? He should!) are stellar and a big part of why those movies are so dope because it's seriously not the acting or the stories...
I also like a lot of porn-soundtracks from the 70's-early 80's but whatever; that's not what this thread is about...

Nine Lives of fritz the cat. The movie may suck compared to the preceding one but it's still pretty cool IMO and the soundtrack is fucking ace with Tom Scott and the LA express (Tom Scott=west coast jazz veteran going commercial; mostly known for the TROY-sample)
